Managers of a "flagship" department store say more than 500 jobs will be created at its new city centre premises.
John Lewis said a store above the redeveloped Birmingham New Street station would offer roles on the shop floor, stock-room and kitchen.
The company said the move represented its "ongoing commitment" to the city.
Due to open in September, the £35m store anchors the Grand Central shopping centre.
When completed, the 250,000 sq ft (23,226 sq m) store will be one of the company's largest outside London.
New Street's £650m face-lift will also feature about 60 shops and restaurants.
